چکیده

We developed a ratiometric method capable of estimating total hemoglobin concentration from optically measured diffuse reflectance spectra. The three isosbestic wavelength ratio pairs that best correlated to total hemoglobin concentration independent of saturation and scattering were 545/390, 452/390, and 529/390 nm. These wavelength pairs were selected using forward Monte Carlo simulations which were used to extract hemoglobin concentration from experimental phantom measurements. Linear regression coefficients from the simulated data were directly applied to the phantom data, by calibrating for instrument throughput using a single phantom. Phantoms with variable scattering and hemoglobin saturation were tested with two different instruments, and the average percent errors between the expected and ratiometrically-extracted hemoglobin concentration were as low as 6.3%. A correlation of r = 0.88 between hemoglobin concentration extracted using the 529/390 nm isosbestic ratio and a scalable inverse Monte Carlo model was achieved for in vivo dysplastic cervical measurements (hemoglobin concentrations have been shown to be diagnostic for the detection of cervical pre-cancer by our group). These results indicate that use of such a simple ratiometric method has the potential to be used in clinical applications where tissue hemoglobin concentrations need to be rapidly quantified in vivo.

Noninvasive monitoring of tissue hemoglobin using UV-VIS diffuse reflectance spectroscopy: a pilot study.

We conducted a pilot study on 10 patients undergoing general surgery to test the feasibility of diffuse reflectance spectroscopy in the visible wavelength range as a noninvasive monitoring tool for blood loss during surgery. Ratios of raw diffuse reflectance at wavelength pairs were tested as a first-pass for estimating hemoglobin concentration. Ratios can be calculated easily and rapidly with ...

Rapid Determination of Oxygen Saturation and Vascularity for Cancer Detection

A rapid heuristic ratiometric analysis for estimating tissue hemoglobin concentration and oxygen saturation from measured tissue diffuse reflectance spectra is presented. The analysis was validated in tissue-mimicking phantoms and applied to clinical measurements in head and neck, cervical and breast tissues. The analysis works in two steps. First, a linear equation that translates the ratio of...

A study on the identification of habitats and determination of sulfur dioxide residue of Radix Astragali by UV-vis-SWNIR diffuse reflectance spectroscopy

The feasibility of identifying habitat of sulfur fumigated Radix Astragali and of determining its sulfur dioxide residue using UV-vis and short wave near-infrared diffuse reflectance spectra coupled with chemometrics was first investigated. The results show that by using UV-vis-SWNIR DRS combined with PCA, the habitat of Radix Astragali is identified definitely, whether it is sulfur-fumigated, ...

Determination of Leaf Relative Water Content of Two Genotypes of Sesame Using Visible and Near- Infrared (VIS/NIR) Spectrometry to Detect Drought Stress

Relative water content (RWC) in plants is one of the most important biochemical parameters and its deficiency limits efficiency of photosynthesis and crop productivity. The scientific reports on using spectroscopy in detecting drought stress for sesame plants are very rare. In this study, the possibility of identifying water stress in two sensitive (Naz-Takshakhe) and resistant (Yekta) genotype...

Diagnosis of breast cancer using diffuse reflectance spectroscopy: Comparison of a Monte Carlo versus partial least squares analysis based feature extraction technique.

BACKGROUND AND OBJECTIVE We explored the use of diffuse reflectance spectroscopy in the ultraviolet-visible (UV-VIS) spectrum for the diagnosis of breast cancer. A physical model (Monte Carlo inverse model) and an empirical model (partial least squares analysis) based approach, were compared for extracting diagnostic features from the diffuse reflectance spectra. STUDY DESIGN/METHODS The phys...
